About	The Somru® C-1 inhibitor(Berinert®) activity assay kit utilizes the ability of the C esterase inhibitor enzyme to inhibit recombinant human complement component C1s and subsequent cleavage of a colorimetric peptide substrate. This assay can be used to measure endogenous C1 inhibitor and for monitoring substitution therapy and steroid therapy in plasma.
Detection method	Colorimetric; Assay can be read as kinetic (ΔA405 nm/minute) or endpoint (A405 nm) method.
Storage	Store the unopened kit at 2-8 °C.
Quality Control Samples (QCs)	Three levels of QCs are included for custom GLP kits. QCs are prepared by fortifying human plasma derived C1-esterase inhibitor in human plasma or serum.
Precision	The precision was determined by analyzing samples prepared at 100nM in 6 replicates on 6 different occasions. Intra-assay precision (coefficient of variation (CV)) ranges from 2% to 7%. Inter-assay precision was at 9%.
Accuracy/ Recovery	100nM C1-inhibitor was fortified in 10 lots of human serum. Recovery ranges are from 91-109% with an average recovery of 104%.
Total Error	Total Error (TE) was calculated by adding the mean %bias with inter-assay variability (%CV). The TE was found to be less than 13%.
Sensitivity	The limit of detection is 0.8 nM.
Dilutional Linearity	Samples can be diluted up to 32 fold. Assay range may be changed to meet intended purpose by changing ratio of sample and C1s. Please contact our technical service team at tec@somrubioscience.com.
Cross-reactivity	No known cross-reactivity exists.
