| About | The Somru® Glucagon ELISA assay employs the quantitative sandwich enzyme immunoassay technique using two monoclonal antibodies specific for pancreatic glucagon. This user-friendly assay design utilizes a pair of antibodies that allows the detection of the whole pancreatic glucagon molecule in biological matrices with minimal or no cross reactivity to other form of glucagon molecules. | ||||||||||
| Detection method | Colorimetric; absorbance at 450 nm with reference absorbance at 650 nm. | ||||||||||

| Quality Control Samples (QCs) | Three levels of QCs are included for custom GLP kits for an additional cost. QCs are prepared by fortifying glucagon in human serum or plasma. | ||||||||||
| Precision | The precision was determined by analyzing samples prepared at 450 pg/mL in 6 replicates on 6 different occasions. Intra-assay precision (coefficient of variation(CV)) ranges from 3% to 7%. Inter-assay precision was at 9%. | ||||||||||
| Accuracy | The accuracy was determined by analyzing three levels of quality control samples in 6 replicates on 6 different occasions. Mean %bias was less than 12%. | ||||||||||
| Total Error | Total Error (TE) was calculated by adding the mean %bias with inter-assay variability (%CV). The TE was found to be less than 15%. | ||||||||||
| Selectivity/ Recovery | The recovery of glucagon was evaluated by fortifying 200 pg/mL of glucagon in various matrices. The recovery ranged from 91% to 107%. | ||||||||||
| Sensitivity | The limit of detection is 11.7 pg/mL. | ||||||||||
| Dilutional Linearity | Dilutional linearity was established up to a dilution of 16 fold. The assay is capable of measuring samples with up to 32000 pg/mL of glucagon. | ||||||||||
